目的 比较锝[???Tc]甲氧基异丁基异腈单光子发射计算机断层扫描/计算机断层扫描(???Tc-MIBI SPECT/CT)双时相显像与颈部超声在继发性甲状旁腺功能亢进症(SHPT)患者术前定位中的诊断价值。方法 回顾性纳入2025年5月至2026年4月于黔东南苗族侗族自治州人民医院行两种影像学检查的疑似SHPT患者54例。以手术病理结果为金标准,计算超声与SPECT/CT在区域层面(共216个区域)的敏感性、特异性、准确性,并比较不同解剖区域的病灶检出率。结果 54例中确诊SHPT 45例(83.33%)。超声诊断敏感性、特异性、准确性分别为70.89%(112/158)、86.21%(50/58)、75.00%(162/216);SPECT/CT分别为92.41%(146/158)、94.83%(55/58)、93.06%(201/216)。SPECT/CT的敏感性和准确性均高于超声(P均<0.001)。SPECT/CT在左上、左下、右上三个区域的检出率均高于超声(P<0.05)。结论 99mTc-MIBI SPECT/CT双时相显像在SHPT术前定位中的敏感性和准确性优于颈部超声,尤其对左上、左下、右上区域病灶检出优势明显,可作为SHPT患者术前定位的重要补充或首选影像学方法。
Objective?To compare the diagnostic value of technetium-99m methoxyisobutylisonitrile single-photon emission computed tomography/computed tomography (99mTc-MIBI SPECT/CT) dual-phase imaging and cervical ultrasound in preoperative localization of secondary hyperparathyroidism (SHPT).?Methods?A total of 54 patients with suspected SHPT who underwent both imaging examinations at Qiandongnan Miao and Dong Autonomous Prefecture People's Hospital from May 2025 to April 2026 were retrospectively enrolled. Using surgical pathology as the gold standard, the sensitivity, specificity, and accuracy of ultrasound and SPECT/CT were calculated at the regional level (216 regions in total), and the detection rates in different anatomical regions were compared.?Results?Among the 54 patients, 45 (83.33%) were confirmed with SHPT. The sensitivity, specificity, and accuracy of ultrasound were 70.89% (112/158), 86.21% (50/58), and 75.00% (162/216), respectively; while those of SPECT/CT were 92.41% (146/158), 94.83% (55/58), and 93.06% (201/216), respectively. The sensitivity and accuracy of SPECT/CT were significantly higher than those of ultrasound (both?P<0.001). The detection rates of SPECT/CT in the upper-left, lower-left, and upper-right regions were significantly higher than those of ultrasound (P<0.05).?Conclusion?99mTc-MIBI SPECT/CT dual-phase imaging demonstrates superior sensitivity and accuracy compared with cervical ultrasound in preoperative localization of SHPT, especially showing significant advantages in detecting lesions in the upper-left, lower-left, and upper-right regions. It can serve as an important supplement or the first-choice imaging modality for preoperative localization in SHPT patients.

目的 观察辅助性T17细胞(Th17)与调节性T细胞(Treg)比值与2型糖尿病(T2DM)患者胰岛素抵抗及胰岛β细胞功能的关系。方法 纳入2022年4月—2023年4月在贵州医科大学第二附属医院内分泌科住院及健康体检人群各100例, 分为糖耐量正常组(NGT组, n=100)和T2DM组(n=100), 分别测定糖化血红蛋白(HbA1c)、空腹血糖(FPG)、甘油三酯(TG)等生化指标, 电化学发光法测定空腹胰岛素(FINS), 稳态模型计算胰岛β细胞功能指数(HOMA-β)、胰岛素抵抗指数(HOMA-IR)及胰岛素敏感指数(HOMA-ISI)。流式细胞术检测Th17、Treg水平。HOMA-IR、HOMA-β和HOMA-ISI的影响因素采用多元线性回归分析。结果 与NGT组相比, T2DM组BMI、FPG、HbA1c、LDL-C 、TG、TC、FINS、HOMA-IR、Th17及Th17/Treg水平均升高(P<0.01), HDL-C、HOMA-β、HOMA-ISI、Treg水平均降低, 且差异有统计学意义(P<0.01)。Th17与BMI(r=0.251, P<0.001)及HOMA-IR(r=0.305, P<0.001)呈正相关; 与HOMA-β(r=-0.204, P<0.001)及HOMA-ISI(r=-0.359, P<0.001)呈负相关。Treg与HOMA-ISI之间呈正相关(r=0.170, P=0.008), 而与HOMA-IR呈负相关(r=-0.153, P=0.017); Th17/Treg与BMI(r=0.332, P<0.001)及HOMA-IR(r=0.374, P<0.001);与HOMA-β(r=-0.249, P<0.001)及HOMA-ISI(r=-0.427, P<0.001)呈负相关。多元线性回归分析显示, Th17/Treg是HOMA-IR(β=5.915)升高及HOMA-ISI(β=-2.557)下降的影响因素(P<0.01)。结论 Th17/Treg可能通过影响胰岛素抵抗、降低胰岛素敏感性参与T2DM的发生。
Objective To explore the relationship among the proportion of helper T17 cells(Th17)to regulatory T cells(Treg), insulin resistance, and the function of islet beta cells.Methods One hundred cases of hospitalized patients and 100 cases of health check-ups people in the Department of Endocrinology of the Second Affiliated Hospital of Guizhou Medical University from April 2022 to April 2023 were included.Patients were divided into normal glucose tolerance group(NGT group, n=100)and type 2 diabetes mellitus group(T2DM group, n=100).The biochemical indexes of HbA1c, fasting blood glucose(FPG), triglyceride(TG)and fasting insulin(FINS)were determined by electrochemiluminescence.Islet beta cell function index(HOMA-β), insulin resistance index(HOMA-IR)and insulin sensitivity index(HOMA-ISI)were calculated in homeostasis model.The levels of Th17 and Treg were detected by flow cytometry.Spearman was used to analyze the correlation between indicators, and multiple linear regression analysis was used to analyze the influencing factors of HOMA-IR, HOMA-β and HOMA-ISI.Results In contrast to the NGT group, the T2DM group exhibited elevated levels of BMI, FPG, HbA1c, LDL-C, TG, TC, FINS, HOMA-IR, Th17 and Th17/Treg, with these variances being signifincantly different(P<0.01).There was a notable reduction in the levels of HDL-C,HOMA-β,HOMA-ISI,Treg,with those changes being significantly different(P<0.01).Th17 was positively correlated with BMI(r=0.251, P<0.001)and HOMA-IR(r=0.305, P<0.001), it was negatively correlated with HOMA-β(r=-0.204, P<0.001)and HOMA-ISI(r=-0.359, P<0.001).Treg was positively correlated with HOMA-ISI(r=0.170, P=0.008), while it was negatively correlated with HOMA-IR(r=-0.153, P=0.017).The ratio of Th17/Treg was positively correlated with BMI(r=0.332, P<0.001)and HOMA-IR(r=0.374, P<0.001), it was negatively correlated with HOMA-β(r=-0.249, P<0.001)and HOMA-ISI(r=-0.427, P<0.001).Multiple linear regression analysis showed that Th17/Treg was an influential factor in the increase of HOMA-IR(β=5.915)and the decrease of HOMA-ISI(β=-2.557)(P<0.01).Conclusions Th17/Treg may be involved in the development of T2DM by affecting insulin resistance and reducing insulin sensitivity.
目的 探讨谷氨酸对HT22细胞线粒体自噬和细胞凋亡的影响,并评估虾青素预处理的保护作用及其分子机制。方法 用谷氨酸及虾青素处理HT22细胞,通过蛋白印迹及聚合酶联反应等评估其对线粒体自噬的影响。结果 谷氨酸处理显著抑制线粒体初级自噬(PINK1、Parkin、pULK1ser555和LC3Ⅱ)和次级自噬(LAMP1和Rab7),上调cleaved Caspase-3的表达(P<0.05)。虾青素预处理减少细胞凋亡,恢复了线粒体自噬,PINK1、Parkin、pULK1ser555和LC3Ⅱ的表达水平上升(分别为2.3倍、2.6倍、83.3%及81.1%)(P<0.05),该作用被自噬抑制剂BafA1阻断。此外,谷氨酸抑制Nrf2核内转移和NLRX1表达,而预处理显著促进Nrf2的核内转移并上调NLRX1,分别上调25.8%、33.2%。生物信息学分析显示NLRX1启动子区域含有3个Nrf2结合位点,提示Nrf2通过调控NLRX1转录活性发挥作用。结论 文章首次揭示虾青素通过Nrf2/NLRX1通路激活线粒体自噬,展现神经保护作用。
Objective To explore the effects of glutamate on mitophagy and apoptosis in HT22 cells and evaluate the protective effects and molecular mechanisms of astaxanthin pretreatment.Methods HT22 cells were treated with glutamate and astaxanthin.The effects on mitophagy were assessed using Western Blot and PCR.Results Glutamate treatment significantly inhibited primary mitophagy(PINK1,Parkin,pULK1ser555 and LC3II)and secondary mitophagy(LAMP1 and Rab7)while upregulating cleaved Caspase-3 expression.Astaxanthin pretreatment notably reduced apoptosis and restored mitophagy,the expression levels of PINK1,Parkin,pULK1Ser555 and LC3II were significantly upregulated(by 2.3-fold,2.6-fold,83.3% and 81.1% respectively,P<0.05),but this effect was blocked by the autophagy inhibitor BafA1.Additionally,glutamate suppressed Nrf2 nuclear translocation and NLRX1 expression,whereas astaxanthin promoted Nrf2 nuclear translocation and increased NLRX1 expression by 25.8% and 33.2%,respectively.Bioinformatics analysis revealed three Nrf2 binding sites in the NLRX1 promoter region,suggesting that Nrf2 may regulate NLRX1 transcriptional activity.Conclusions The study demonstrates that astaxanthin exhibited potential neuroprotective effect by activating mitophagy through the Nrf2/NLRX1 pathway.
目的 研究SOCS6/STAT6通路在前列腺细胞炎性增殖作用中的调控作用。方法 使用人前列腺细胞株RWPE-1建立炎症模型,将细胞分为对照(CON)组和炎症刺激(INF)组,后者通过添加脂多糖(LPS)模拟炎症环境。采用ELISA检测白细胞介素-1β(IL-1β)-1β、白细胞介素-6(IL-6)和白细胞介素-8(IL-8)表达水平,蛋白免疫印迹法检测细胞因子信号抑制物-6(SOCS6)、信号转导和转录激活因子-6(STAT6)及磷酸化STAT6蛋白的表达水平。结果 经过LPS处理后,RWPE-1细胞中的SOCS6蛋白表达水平显著下降(P<0.01),而磷酸化STAT6表达水平上升(P<0.01)。结论 SOCS6/STAT6通路可能通过调节炎症环境下STAT6的磷酸化水平,参与调节前列腺细胞的炎性增殖作用。
Objective To explore the regulatory role of SOCS6/STAT6 pathway in the inflammatory proliferation of prostate cells.Methods The human prostate cell line RWPE-1 was used to establish an inflammation model.Cells were divided into a control(CON)group and an inflammation-stimulated(INF)group,with the latter subjected to lipopolysaccharide(LPS)treatment to simulate an inflammatory environment.The expression levels of interleukin(IL)-1β、IL-6 and IL-8 were detected by ELISA,and the expression levels of suppressor of cytokine signaling 6(SOCS6),signal transducer and activator oftranscription-6,(STAT6),and phosphorylated STAT6 proteins were detected by Western blot.Results The results showed that after LPS treatment,the expression of SOCS6 protein in RWPE-1 cells significantly decreased,while the expression of phosphorylated STAT6 increased.Conclusions The SOCS6/STAT6 pathway may be involved in regulating the inflammatory proliferation of prostate cells by modulating the phosphorylation level of STAT6 under inflammatory conditions.
